Product Description

ESOLIN-D SUSTAINED RELEASE CAPSULES
Esomeprazole and Domperidone Sustained Release Capsules

Dosage Form: Sustained Release Capsules

Family: Proton Pump Inhibitor (PPI) and Prokinetic Agent Combination

Indications: ESOLIN-D is used for the management of gastroesophageal reflux disease (GERD), acid reflux, heartburn, and selected acid-related gastrointestinal disorders associated with nausea or delayed gastric emptying, where both ingredients are clinically appropriate.

Mode of Action: Esomeprazole suppresses gastric acid secretion by inhibiting the proton pump (H+/K+-ATPase) in the stomach's parietal cells. Domperidone blocks peripheral dopamine D2 receptors, helping improve upper gastrointestinal motility and gastric emptying while reducing nausea and vomiting.

Clinical Benefits: The combination helps reduce excessive stomach acid and supports the management of acid reflux symptoms. Domperidone may improve gastric emptying and relieve associated nausea, bloating, and upper abdominal discomfort in appropriately selected patients.

Therapeutic Benefits: ESOLIN-D combines acid suppression with a prokinetic effect, which may be beneficial for selected patients who experience reflux symptoms alongside nausea or delayed gastric emptying. Treatment should be tailored to the patient's diagnosis and clinical needs.

Dosage and Administration: Take orally according to the prescription and approved product instructions. The dose and timing depend on the strength, formulation, and clinical condition. Swallow the capsule whole with water; do not crush or chew sustained-release contents. Do not exceed the prescribed dose or duration.

Side Effects: Possible side effects include headache, abdominal pain, diarrhea, constipation, nausea, dry mouth, and dizziness. Domperidone may also cause elevated prolactin levels, which can occasionally lead to breast tenderness or milk secretion.

Serious Adverse Effects: Domperidone has been associated with QT prolongation, serious abnormal heart rhythms, and a small increased risk of sudden cardiac death in susceptible patients. Seek urgent medical attention for fainting, palpitations, or an irregular heartbeat.

Contraindications: Do not use in patients with known hypersensitivity to either ingredient, gastrointestinal bleeding or obstruction, or prolactinoma. Domperidone is unsuitable for patients with certain cardiac conditions, significant electrolyte disturbances, or concurrent use of medicines that strongly inhibit CYP3A4 or prolong the QT interval.

Precautions: Use the lowest effective domperidone dose for the shortest appropriate duration. Particular caution is required in older adults and patients with heart disease, liver impairment, or electrolyte abnormalities. Review the need for treatment regularly.

Drug Interactions: Esomeprazole may interact with medicines such as clopidogrel and certain drugs whose absorption depends on stomach acidity. Domperidone may interact with macrolide antibiotics, azole antifungals, and other medicines that prolong the QT interval or inhibit CYP3A4. A healthcare professional should review all concurrent medicines.

Use During Pregnancy: Use only when considered necessary by a qualified healthcare professional after assessing the potential benefits and risks of both ingredients.

Use During Breastfeeding: Domperidone passes into breast milk and may affect the infant. Use during breastfeeding should be assessed by a healthcare professional, considering the safety of both ingredients and available alternatives.

Storage: Store according to the manufacturer's approved product instructions. Protect from excessive heat, moisture, and direct sunlight. Keep out of the reach of children.
